Prostaglandin-Endoperoxid-Synthase 2, auch bekannt als Cyclooxygenase-2 oder einfach COX-2, ist ein Enzym, das beim Menschen durch das PTGS2-Gen kodiert wird. Es gibt zwei Isoenzyme der COX, die durch unterschiedliche Genprodukte kodiert werden: eine konstitutive COX-1 (dieses Enzym) und eine induzierbare COX-2, die sich in ihrer Expressionsregulation und Gewebeverteilung unterscheiden. Die Expression dieser beiden Transkripte wird durch relevante Zytokine und Wachstumsfaktoren differentiell reguliert. Eine Spleißvariante der COX-1, genannt COX-3, wurde im ZNS von Hunden identifiziert, führt aber beim Menschen nicht zu einem funktionellen Protein. Zwei kleinere COX-1-abgeleitete Proteine (die partiellen COX-1-Proteine PCOX-1A und PCOX-1B) wurden ebenfalls entdeckt, aber ihre genauen Rollen müssen noch beschrieben werden.
