MAP2 ist das wichtigste Mikrotubuli-assoziierte Protein des Gehirngewebes. Es gibt drei Formen von MAP2; zwei haben eine ähnliche Größe mit apparenten Molekulargewichten von 280 kDa (MAP2a und MAP2b) und die dritte hat ein geringeres Molekulargewicht von 70 kDa (MAP2c). Im Gehirn neugeborener Ratten sind MAP2b und MAP2c vorhanden, während MAP2a fehlt. Zwischen dem 10. und 20. postnatalen Tag erscheint MAP2a. Gleichzeitig sinkt der Spiegel von MAP2c um das 10-fache. Diese Veränderung tritt in der Zeit auf, in der das Dendritenwachstum abgeschlossen ist und die Neuronen ihre reife Morphologie erreicht haben. MAP2 wird im Gehirn alternder Ratten durch eine Cathepsin-D-ähnliche Protease abgebaut. Es gibt Hinweise darauf, dass MAP2 in einigen Neuronenarten in höheren Mengen exprimiert wird als in anderen. MAP2 ist bekannt dafür, die Mikrotubuli-Assemblierung zu fördern und Seitenarme an Mikrotubuli zu bilden. Es interagiert auch mit Neurofilamenten, Aktin und anderen Elementen des Zytoskeletts.
