Das PIGR (Polymeric Immunoglobulin Receptor) ELISA Kit für Rinder von ELK Biotechnology ist ein ELISA Kit zur Messung von PIGR in der Forschung.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem Antikörper vorbeschichtet, der spezifisch für Rinder-R ist. Standards oder Proben werden zu den entsprechenden Mikrotiterplattenvertiefungen hinzugefügt, gefolgt von einem Biotin-konjugierten Antikörper, der spezifisch für Rinder-R ist. Anschließend wird Avidin, konjugiert an Meerrettichperoxidase (HRP), zu jeder Mikrotiterplattenvertiefung hinzugefügt und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur diejenigen Vertiefungen, die Rinder-R, Biotin-konjugierten Antikörper und Enzym-konjugiertes Avidin enthalten, eine Farbänderung. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet, und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von Rinder-R in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
