Das Maus-MHCDQb1(Major Histocompatibility Complex Class II DQ Beta 1) ELISA Kit ist ein ELISA-Kit von ELK Biotechnology zur Forschungsmessung von MHCDQb1.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem Antikörper vorbeschichtet, der spezifisch für Maus-MHCDQb1 ist. Standards oder Proben werden dann zusammen mit einem Biotin-konjugierten Antikörper, der spezifisch für Maus-MHCDQb1 ist, in die entsprechenden Mikrotiterplattenvertiefungen gegeben. Als Nächstes wird Avidin, das an Meerrettichperoxidase (HRP) konjugiert ist, zu jeder Mikrotiterplattenvertiefung gegeben und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur diejenigen Vertiefungen eine Farbänderung, die Maus-MHCDQb1, Biotin-konjugierten Antikörper und Enzym-konjugiertes Avidin enthalten. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet, und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von Maus-MHCDQb1 in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
