Das Maus TPH1 (Tryptophan-Hydroxylase 1) ELISA Kit ist ein ELISA-Kit von ELK Biotechnology zur Erforschung und Messung von TPH1.
Das in diesem Kit angewendete Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem Antikörper vorbeschichtet, der spezifisch für Maus-TPH1 ist. Standards oder Proben werden in die entsprechenden Vertiefungen der Mikrotiterplatte gegeben, dann mit einem biotin-konjugierten Antikörper, der spezifisch für Maus-TPH1 ist. Anschließend wird Avidin, konjugiert mit Meerrettichperoxidase (HRP), zu jeder Mikrotiterplattenvertiefung hinzugefügt und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur diejenigen Vertiefungen, die Maus-TPH1, biotin-konjugierten Antikörper und enzymkonjugiertes Avidin enthalten, eine Farbänderung. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von Maus-TPH1 in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
