Das Ratten-ACO1(Aconitase 1) ELISA Kit ist ein ELK Biotechnology ELISA Kit für die Forschungsmessung von ACO1.
Das in diesem Kit angewandte Testprinzip ist ein Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem Antikörper vorbeschichtet, der spezifisch für Ratten-ACO1 ist. Standards oder Proben werden in die entsprechenden Mikrotiterplattenvertiefungen gegeben, dann mit einem biotin-konjugierten Antikörper, der spezifisch für Ratten-ACO1 ist. Als Nächstes wird Avidin, konjugiert mit Meerrettichperoxidase (HRP), zu jeder Mikrotiterplattenvertiefung gegeben und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur jene Vertiefungen, die Ratten-ACO1, einen biotin-konjugierten Antikörper und enzymkonjugiertes Avidin enthalten, eine Farbänderung. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von Ratten-ACO1 in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.

Aconitase 1 (ACO1), also known as cytoplasmic aconitate hydratase and iron regulatory protein 1 (IRP1), is a bifunctional human protein linking citrate metabolism with iron-responsive post-transcriptional regulation.
Biological function: ACO1 is a bifunctional cytosolic protein. When assembled with a [4Fe-4S] cluster it acts as aconitase, catalysing the reversible interconversion of citrate and isocitrate through cis-aconitate. Under iron-limiting conditions, loss of the cluster enables ACO1/IRP1 to bind iron-responsive elements in target mRNAs and regulate proteins involved in cellular iron uptake, storage and use.
