Das Ratten-PLA2G2A- (Phospholipase A2, Gruppe IIA) ELISA-Kit ist ein ELK Biotechnology ELISA-Kit zur Forschungsmessung von PLA2G2A.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte ist mit einem Antikörper vorbeschichtet, der spezifisch für Ratten-PLA2G2A ist. Standards oder Proben werden den entsprechenden Vertiefungen der Mikrotiterplatte hinzugefügt, gefolgt von einem Biotin-konjugierten Antikörper, der spezifisch für Ratten-PLA2G2A ist. Anschließend wird Avidin, das an Meerrettichperoxidase (HRP) konjugiert ist, zu jeder Mikrotiterplattenvertiefung gegeben und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur jene Vertiefungen, die Ratten-PLA2G2A, Biotin-konjugierten Antikörper und Enzym-konjugiertes Avidin enthalten, eine Farbveränderung. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von Ratten-PLA2G2A in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
