Das Ratten-SCGN(Secretagogin)-ELISA-Kit ist ein ELK Biotechnology ELISA-Kit zur Messung von SCGN in der Forschung.
Dieser Assay verwendet die kompetitive Enzymimmunoassay-Technik mit kompetitiver Inhibition. Die in diesem Kit enthaltene Mikrotiterplatte ist mit Ratten-SCGN vorbeschichtet. Standards oder Proben werden in die entsprechenden Mikrotiterplattenvertiefungen gegeben, dann mit einem biotin-konjugierten Antikörper, der spezifisch für Ratten-SCGN ist. Anschließend wird Avidin, das an Meerrettichperoxidase (HRP) konjugiert ist, zu jeder Mikrotiterplattenvertiefung hinzugefügt und inkubiert. Danach wird TMB-Substratlösung hinzugefügt. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von Ratten-SCGN in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
