Das Maus ADAM17 (A Disintegrin And Metalloprotease 17) ELISA Kit ist ein ELISA Kit von ELK Biotechnology für die Forschungsmessung von ADAM17.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem Antikörper vorbeschichtet, der spezifisch für Maus ADAM17 ist. Standards oder Proben werden zu den entsprechenden Mikrotiterplattenvertiefungen hinzugefügt, gefolgt von einem biotin-konjugierten Antikörper, der spezifisch für Maus ADAM17 ist. Als Nächstes wird Avidin, das an Meerrettichperoxidase (HRP) konjugiert ist, zu jeder Mikrotiterplattenvertiefung hinzugefügt und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur jene Vertiefungen, die Maus ADAM17, den biotin-konjugierten Antikörper und das enzymkonjugierte Avidin enthalten, eine Farbveränderung. Die Enzym-Substrat-Reaktion wird durch Zugabe einer Schwefelsäurelösung beendet und der Farbwechsel wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von Maus ADAM17 in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
