Der Human MFN2 (Mitofusin 2) Microsample ELISA Kit ist ein ELK Biotechnology ELISA Kit für die Forschungsmessung von MFN2.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem Antikörper vorbeschichtet, der spezifisch für humanes MFN2 ist. Standards oder Proben werden in die entsprechenden Vertiefungen der Mikrotiterplatte gegeben, anschließend ein Biotin-konjugierter Antikörper, der spezifisch für humanes MFN2 ist. Als Nächstes wird Avidin, konjugiert an Meerrettichperoxidase (HRP), zu jeder Mikrotiterplattenvertiefung gegeben und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur jene Vertiefungen eine Farbveränderung, die humanes MFN2, den Biotin-konjugierten Antikörper und das Enzym-konjugierte Avidin enthalten. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und die Farbveränderung spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von humanem MFN2 in den Proben wird dann durch Vergleich der OD-Werte der Proben mit der Standardkurve bestimmt.
