Der Human ACTb (Aktin Beta) Microsample ELISA Kit ist ein ELK Biotechnology ELISA Kit zur Messung von ACTb in der Forschung.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte ist mit einem Antikörper vorbeschichtet, der spezifisch für humanes ACTb ist. Standards oder Proben werden in die entsprechenden Vertiefungen der Mikrotiterplatte gegeben, danach ein biotin-konjugierter Antikörper, der spezifisch für humanes ACTb ist. Als Nächstes wird Avidin, konjugiert an Meerrettichperoxidase (HRP), zu jeder Mikrotiterplattenvertiefung hinzugefügt und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur diejenigen Vertiefungen eine Farbänderung, die humanes ACTb, den biotin-konjugierten Antikörper und das enzymkonjugierte Avidin enthalten. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet, und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von humanem ACTb in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.

Biological function: ACTB is a highly conserved actin that polymerises from monomeric G-actin into F-actin filaments and cross-linked cytoskeletal networks.
