Das Humane TCHH (Trichohyalin) ELISA Kit ist ein ELK Biotechnology ELISA Kit für die Forschungsmessung von TCHH.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte ist mit einem Antikörper beschichtet, der spezifisch für humanes TCHH ist. Standards oder Proben werden in die entsprechenden Mikrotiterplattenvertiefungen gegeben, dann mit einem Biotin-konjugierten Antikörper, der spezifisch für humanes TCHH ist. Als Nächstes wird Avidin, das an Meerrettichperoxidase (HRP) konjugiert ist, zu jeder Mikrotiterplattenvertiefung gegeben und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur diejenigen Vertiefungen eine Farbänderung, die humanes TCHH, Biotin-konjugierten Antikörper und Enzym-konjugiertes Avidin enthalten. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von humanem TCHH in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
