Der Human-NFkB2-(Nuclear Factor Kappa B2)-Mikroproben-ELISA-Kit ist ein ELK Biotechnology ELISA-Kit zur Forschungsanalyse von NFkB2.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem für humanes NFkB2 spezifischen Antikörper vorbeschichtet. Standards oder Proben werden in die entsprechenden Mikrotiterplattenvertiefungen gegeben, dann mit einem biotin-konjugierten Antikörper, der spezifisch für humanes NFkB2 ist. Anschließend wird Avidin, das an Meerrettichperoxidase (HRP) konjugiert ist, zu jeder Mikrotiterplattenvertiefung gegeben und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur jene Vertiefungen, die humanes NFkB2, biotin-konjugierten Antikörper und enzymkonjugiertes Avidin enthalten, eine Farbänderung. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet, und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration des humanen NFkB2 in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
