Das Humane PDIA4(Protein Disulfide Isomerase A4) ELISA Kit ist ein ELK Biotechnology ELISA Kit für die Forschungsmessung von PDIA4.
Das in diesem Kit angewandte Testprinzip ist ein Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem Antikörper vorbeschichtet, der spezifisch für humanes PDIA4 ist. Standards oder Proben werden in die entsprechenden Vertiefungen der Mikrotiterplatte gegeben, gefolgt von einem Biotin-konjugierten Antikörper, der spezifisch für humanes PDIA4 ist. Anschließend wird Avidin, konjugiert an Meerrettichperoxidase (HRP), zu jeder Mikrotiterplattenvertiefung gegeben und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur jene Vertiefungen, die humanes PDIA4, Biotin-konjugierten Antikörper und Enzym-konjugiertes Avidin enthalten, eine Farbänderung. Die Enzym-Substrat-Reaktion wird durch Zugabe einer Schwefelsäurelösung beendet und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450nm ± 10nm gemessen. Die Konzentration des humanen PDIA4 in den Proben wird dann durch Vergleich der OD-Werte der Proben mit der Standardkurve bestimmt.
