Der ELISA-Kit für humanes 5HTR1A (5-Hydroxytryptamin-Rezeptor 1A) ist ein ELISA-Kit von ELK Biotechnology zur Forschungsmessung von 5HTR1A.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem für humanes 5HTR1A spezifischen Antikörper vorbeschichtet. Standards oder Proben werden in die entsprechenden Mikrotiterplattenvertiefungen gegeben, danach ein biotin-konjugierter Antikörper, der spezifisch für humanes 5HTR1A ist. Als Nächstes wird mit Meerrettichperoxidase (HRP) konjugiertes Avidin zu jeder Mikrotiterplattenvertiefung gegeben und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur diejenigen Vertiefungen eine Farbänderung, die humanes 5HTR1A, biotin-konjugierten Antikörper und enzymkonjugiertes Avidin enthalten. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von humanem 5HTR1A in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
