Der Human KRT7 (Keratin 7) Microsample ELISA Kit ist ein ELK Biotechnology ELISA Kit zur Messung von KRT7 zu Forschungszwecken.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem Antikörper vorbeschichtet, der spezifisch für humanes KRT7 ist. Standards oder Proben werden in die entsprechenden Vertiefungen der Mikrotiterplatte gegeben und anschließend mit einem biotin-konjugierten Antikörper, der spezifisch für humanes KRT7 ist. Anschließend wird Avidin, konjugiert mit Meerrettichperoxidase (HRP), zu jeder Mikrotiterplattenvertiefung gegeben und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur die Vertiefungen, die humanes KRT7, biotin-konjugierten Antikörper und enzymkonjugiertes Avidin enthalten, eine Farbänderung. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von humanem KRT7 in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
