Das Rat S100A8 (S100 Calcium Binding Protein A8) Mikrosample ELISA Kit ist ein ELK Biotechnology ELISA Kit zur Messung von S100A8 für Forschungszwecke.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte ist mit einem Antikörper vorbeschichtet, der spezifisch für Ratten-S100A8 ist. Standards oder Proben werden zu den entsprechenden Vertiefungen der Mikrotiterplatte hinzugefügt, dann mit einem Biotin-konjugierten Antikörper, der spezifisch für Ratten-S100A8 ist. Anschließend wird Avidin, konjugiert an Meerrettichperoxidase (HRP), zu jeder Mikrotiterplattenvertiefung hinzugefügt und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur jene Vertiefungen, die Ratten-S100A8, biotin-konjugierten Antikörper und Enzym-konjugiertes Avidin enthalten, eine Farbänderung. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von Ratten-S100A8 in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
