Das Human KRT14 (Keratin 14) ELISA Kit ist ein ELK Biotechnology ELISA Kit zur Forschungs-Messung von KRT14.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte ist mit einem spezifischen Antikörper gegen humanes KRT14 vorbeschichtet. Standards oder Proben werden in die entsprechenden Vertiefungen der Mikrotiterplatte gegeben, danach ein Biotin-konjugierter Antikörper, der spezifisch für humanes KRT14 ist. Anschließend wird Avidin, konjugiert mit Meerrettichperoxidase (HRP), zu jeder Mikrotiterplattenvertiefung hinzugefügt und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur die Vertiefungen, die humanes KRT14, Biotin-konjugierten Antikörper und Enzym-konjugiertes Avidin enthalten, einen Farbwechsel. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und der Farbwechsel wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von humanem KRT14 in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
