Das Human MMP25 (Matrix Metalloproteinase 25) Mikrosamplen-ELISA-Kit ist ein ELISA-Kit von ELK Biotechnology zur Erforschung und Messung von MMP25.
Das in diesem Kit angewendete Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte ist mit einem für Human MMP25 spezifischen Antikörper vorbeschichtet. Standards oder Proben werden in die entsprechenden Mikrotiterplattenvertiefungen gegeben, danach ein biotin-konjugierter Antikörper, der spezifisch für Human MMP25 ist. Als Nächstes wird Avidin, das an Meerrettichperoxidase (HRP) konjugiert ist, zu jeder Mikrotiterplattenvertiefung gegeben und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur jene Vertiefungen, die Human MMP25, den biotin-konjugierten Antikörper und das enzymkonjugierte Avidin enthalten, eine Farbveränderung. Die Enzym-Substrat-Reaktion wird durch Zugabe einer Schwefelsäurelösung beendet und die Farbveränderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von Human MMP25 in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
