Der ELISA-Kit für humanes ADAMTS4 (A Disintegrin And Metalloproteinase With Thrombospondin 4) ist ein ELISA-Kit von ELK Biotechnology zur Forschungsmessung von ADAMTS4.
Das in diesem Kit angewandte Testprinzip ist ein Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem Antikörper vorbeschichtet, der spezifisch für humanes ADAMTS4 ist. Standards oder Proben werden in die entsprechenden Mikrotiterplattenvertiefungen gegeben, gefolgt von einem biotin-konjugierten Antikörper, der spezifisch für humanes ADAMTS4 ist. Anschließend wird Avidin, das an Meerrettichperoxidase (HRP) konjugiert ist, zu jeder Mikrotiterplattenvertiefung gegeben und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur die Vertiefungen, die humanes ADAMTS4, biotin-konjugierten Antikörper und enzymkonjugiertes Avidin enthalten, eine Farbänderung. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von humanem ADAMTS4 in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
