Der Humane PCDH15 (Protocadherin 15) Mikrosample ELISA Kit ist ein ELK Biotechnology ELISA Kit für die Forschungsbestimmung von PCDH15.
Das in diesem Kit angewandte Testprinzip ist ein Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem spezifischen Antikörper gegen humanes PCDH15 vorbeschichtet. Standards oder Proben werden den entsprechenden Vertiefungen der Mikrotiterplatte hinzugefügt, anschließend ein Biotin-konjugierter Antikörper, der spezifisch für humanes PCDH15 ist. Als Nächstes wird Avidin, konjugiert an Meerrettichperoxidase (HRP), zu jeder Mikrotiterplattenvertiefung hinzugefügt und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur jene Vertiefungen, die humanes PCDH15, biotin-konjugierten Antikörper und enzymkonjugiertes Avidin enthalten, eine Farbveränderung. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und der Farbwechsel wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von humanem PCDH15 in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
