Das Human-FGg (Fibrinogen Gamma) Mikrosampel-ELISA-Kit ist ein ELISA-Kit von ELK Biotechnology zur Forschungsmessung von FGg.
Das in diesem Kit angewendete Testprinzip ist ein Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem für humanes FGg spezifischen Antikörper vorbeschichtet. Standards oder Proben werden in die entsprechenden Vertiefungen der Mikrotiterplatte gegeben, anschließend wird ein biotinylierter Antikörper, der spezifisch für humanes FGg ist, hinzugefügt. Als Nächstes wird Avidin, konjugiert an Meerrettichperoxidase (HRP), zu jeder Mikrotiterplattenvertiefung gegeben und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur die Vertiefungen, die humanes FGg, biotinylierten Antikörper und enzymkonjugiertes Avidin enthalten, eine Farbänderung. Die Enzym-Substrat-Reaktion wird durch Zugabe einer Schwefelsäurelösung beendet und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von humanem FGg in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
