Das Human NT5E (5'-Nucleotidase, Ekto) ELISA Kit ist ein ELK Biotechnology ELISA Kit für die Forschungsmessung von NT5E.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem Antikörper vorbeschichtet, der spezifisch für humanes NT5E ist. Standards oder Proben werden dann zusammen mit einem biotin-konjugierten Antikörper, der spezifisch für humanes NT5E ist, in die entsprechenden Vertiefungen der Mikrotiterplatte gegeben. Anschließend wird Avidin, konjugiert mit Meerrettichperoxidase (HRP), zu jeder Mikrotiterplattenvertiefung gegeben und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur jene Vertiefungen eine Farbänderung, die humanes NT5E, biotin-konjugierten Antikörper und enzymkonjugiertes Avidin enthalten. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet, und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von humanem NT5E in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
