Der Humane IAP (Integrin Associated Protein) Mikrosample ELISA Kit ist ein ELK Biotechnology ELISA Kit zur Messung von IAP für Forschungszwecke.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte ist mit einem spezifischen Antikörper für humanes IAP vorbeschichtet. Standards oder Proben werden in die entsprechenden Vertiefungen der Mikrotiterplatte gegeben, dann ein Biotin-konjugierter Antikörper, der spezifisch für humanes IAP ist. Anschließend wird Avidin, das an Meerrettichperoxidase (HRP) konjugiert ist, zu jeder Mikrotiterplattenvertiefung gegeben und inkubiert. Nachdem die TMB-Substratlösung hinzugefügt wurde, zeigen nur die Vertiefungen, die humanes IAP, biotin-konjugierten Antikörper und enzymkonjugiertes Avidin enthalten, eine Farbänderung. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration des humanen IAP in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
