Der Human a1BG(Alpha-1-B-Glykoprotein) ELISA Kit ist ein ELK Biotechnology ELISA Kit für die Forschungs-Messung von a1BG.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem Antikörper vorbeschichtet, der spezifisch für humanes a1BG ist. Standards oder Proben werden dann zusammen mit einem Biotin-konjugierten Antikörper, der spezifisch für humanes a1BG ist, in die entsprechenden Mikrotiterplattenvertiefungen gegeben. Anschließend wird Avidin, konjugiert an Meerrettichperoxidase (HRP), zu jeder Mikrotiterplattenvertiefung gegeben und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur jene Vertiefungen eine Farbänderung, die humanes a1BG, Biotin-konjugierten Antikörper und Enzym-konjugiertes Avidin enthalten. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von humanem a1BG in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
