Das Human MUTYH (MutY Homolog) ELISA Kit ist ein ELISA Kit von ELK Biotechnology zur Erforschung von MUTYH.
Das in diesem Kit angewandte Testprinzip ist ein Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem spezifischen Antikörper gegen humanes MUTYH vorbeschichtet. Standards oder Proben werden zu den entsprechenden Vertiefungen der Mikrotiterplatte hinzugefügt, anschließend ein biotin-konjugierter Antikörper, der spezifisch für humanes MUTYH ist. Als Nächstes wird Avidin, konjugiert an Meerrettichperoxidase (HRP), zu jeder Mikrotiterplattenvertiefung hinzugefügt und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur jene Vertiefungen, die humanes MUTYH, einen biotin-konjugierten Antikörper und enzymkonjugiertes Avidin enthalten, einen Farbwechsel. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und der Farbwechsel spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von humanem MUTYH in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
