Das Human-HIF2a(Hypoxie-induzierbarer Faktor 2 Alpha)-ELISA-Kit ist ein ELK-Biotechnologie-ELISA-Kit zur Forschungsanalyse von HIF2a.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem für humanes HIF2a spezifischen Antikörper vorbeschichtet. Standards oder Proben werden zu den entsprechenden Mikrotiterplattenvertiefungen gegeben, danach ein Biotin-konjugierter Antikörper, der für humanes HIF2a spezifisch ist. Anschließend wird Avidin, konjugiert mit Meerrettichperoxidase (HRP), zu jeder Mikrotiterplattenvertiefung gegeben und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur die Vertiefungen, die humanes HIF2a, einen Biotin-konjugierten Antikörper und ein Enzym-konjugiertes Avidin enthalten, eine Farbänderung. Die Enzym-Substrat-Reaktion wird durch Zugabe einer Schwefelsäurelösung beendet und die Farbänderung spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von humanem HIF2a in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
