Das Rat MUC1 (Mucin 1) Microsample ELISA Kit ist ein ELISA Kit von ELK Biotechnology zur Erforschung und Messung von MUC1.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem Ratten-MUC1-spezifischen Antikörper vorbeschichtet. Standards oder Proben werden in die entsprechenden Vertiefungen der Mikrotiterplatte gegeben, dann mit einem Biotin-konjugierten Ratten-MUC1-spezifischen Antikörper. Anschließend wird Avidin, konjugiert mit Meerrettichperoxidase (HRP), zu jeder Mikrotiterplattenvertiefung gegeben und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur jene Vertiefungen, die Ratten-MUC1, Biotin-konjugierten Antikörper und Enzym-konjugiertes Avidin enthalten, eine Farbänderung. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von Ratten-MUC1 in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
