Technische Details

Workflow Die Reverse Transkription und die Real-Time-PCR werden in einem Schritt durchgeführt. FAM dient zum Nachweis von LCV, Cy5 als interne Positivkontrolle. Die Reaktion wird bei 60 °C durchgeführt. Das Gesamtvolumen der Reaktion beträgt 25 µL. Den folgenden Standard-PCR-Zyklus verwenden: 50 °C für 5 Minuten, 95 °C für 1 Minute; dann 40 Zyklen von 95 °C für 5 Sekunden, 60 °C für 10 Sekunden; gefolgt von einem Schmelzkurvenprogramm von 95 °C für 1 Minute, 95 °C für 5 Sekunden, 60 °C für 10 Sekunden.
