Das VWF-Gen kodiert den Von-Willebrand-Faktor (VWF), ein großes multimeres Glykoprotein, das eine zentrale Rolle im Blutgerinnungssystem spielt, indem es sowohl ein wichtiger Mediator der Plättchen-Gefäßwand-Interaktion und Plättchenadhäsion als auch ein Träger für den Gerinnungsfaktor VIII ist. VWF, das aus den Weibel-Palade-Körpern der Endothelzellen freigesetzt wird, bindet besonders stark an die extrazelluläre Matrix. Ein VWF-Mangel oder eine VWF-Dysfunktion (Von-Willebrand-Krankheit) führt zu einer Blutungsneigung, die in Geweben mit hohem Blutfluss-Scherdruck in engen Gefäßen am deutlichsten ist.
