Das kodierte Protein ist eine von mehreren verschiedenen Arten von Alpha-Untereinheiten und bildet den Hauptteil der katalytischen Stelle des aktiven Enzyms. In Kollagen und verwandten Proteinen katalysiert die Prolyl-4-Hydroxylase die Bildung von 4-Hydroxyprolin, das für die korrekte dreidimensionale Faltung neu synthetisierter Prokollagenketten unerlässlich ist. Alternativ gespleißte Transkriptvarianten, die verschiedene Isoformen kodieren, wurden beschrieben.
