HDAC3 (HISTON-DEACETYLASE 3) ist ein Mitglied der Histon-Deacetylase/Acuc/Apha-Proteinfamilie, ein Enzym, das beim Menschen durch das HDAC3-Gen kodiert wird. Das HDAC3-Gen ist auf 5q31.3 kartiert. HDAC3 besitzt Histon-Deacetylase-Aktivität und unterdrückt die Transkription, wenn es an einen Promotor gebunden ist. Es kann an der Regulation der Transkription durch seine Bindung an den Zinkfinger-Transkriptionsfaktor YY1 beteiligt sein. Das Protein kann auch die p53-Funktion herunterregulieren und somit Zellwachstum und Apoptose modulieren. Und dieses Gen wird als potenzielles Tumorsuppressorgen angesehen. HDAC3 hat einen offenen Leserahmen von 428 Aminosäuren und teilt 53 % Aminosäureidentität mit HDAC1 und 52 % mit HDAC2. Die katalytische Domäne von HDAC4 interagiert mit HDAC3 über den transkriptionellen Korepressor NCOR2. Alle experimentellen Bedingungen, die zur Unterdrückung der HDAC4-Bindung an NCOR2 und an HDAC3 führten, führten zum Verlust der enzymatischen Aktivität, die mit HDAC4 verbunden ist. Die Rekrutierung von HDAC3 in das Genom zeigt einen zirkadianen Rhythmus in der Mausleber.
