FLT3LG (FMS-Related Tyrosine Kinase 3 Ligand), auch FLT3 LIGAND, FL oder FLT3L genannt, ist ein Protein, das beim Menschen vom FLT3LG-Gen kodiert wird. FLT3LG steuert die Entwicklung von DCs und ist besonders wichtig für plasmazytoide DCs und CD8-positive klassische DCs sowie deren CD103-positive Gewebe-Gegenstücke. Flt3-Ligand (FL) ist ein hämatopoetisches Zytokin mit vier helikalen Bündeln. Es ist strukturell homolog zum Stammzellfaktor (SCF) und zum Kolonie-stimulierenden Faktor 1 (CSF-1). In Synergie mit anderen Wachstumsfaktoren stimuliert der Flt3-Ligand die Proliferation und Differenzierung verschiedener Blutzellvorläufer. Lyman et al. (1993) fanden heraus, dass der Flt3-Ligand die Proliferation hämatopoetischer Vorläuferzellen stimulierte, die aus der fetalen Leber von Mäusen oder dem Knochenmark erwachsener Mäuse isoliert wurden. Hannum et al. (1994) kamen zu dem Schluss, dass FL die Reaktion von Stamm- und primitiven Vorläuferzellen auf andere Wachstumsfaktoren verstärkt, um alle myeloischen Linien außer Erythrozyten zu erzeugen. Assays von C-terminal verkürzten FL-Proteinen bestätigten, dass die N-terminale Hälfte die FL-Aktivität vermittelte. Die Depletion des Pi3k-Mtor-Negativregulators Pten erleichterte die Flt3l-gesteuerte DC-Entwicklung in Kultur.
