CD40 ist ein Zelloberflächenrezeptor, der auf der Oberfläche aller reifen B-Zellen exprimiert wird und auch auf Monozyten, dendritischen Zellen und dem Thymusepithel vorkommt. CD40 ist ein Mitglied der TNF-Rezeptor-Superfamilie (Tumornekrosefaktor) und der Rezeptor für den CD40-Liganden. Der CD40-Ligand (CD40L, CD154, gp39 und TRAM) gehört zur TNF-Genfamilie und wird breiter als CD40 hauptsächlich auf aktivierten CD4+-T-Zellen exprimiert. Die Aktivierung von CD40 hat sich auch als wachstumshemmend für bestimmte B-Zell-Lymphome erwiesen und induziert den Tod transformierter Zellen mesenchymalen oder epithelialen Ursprungs.
