Beclin-1, auch bekannt als ATG6 oder VPS30, ist ein Protein, das beim Menschen durch das BECN1-Gen kodiert wird. Beclin-1 und sein Bindungspartner, die Klasse III Phosphoinositid-3-Kinase (PI3K), auch Vps34 genannt, sind für die Initiierung der Autophagosombildung bei der Autophagie erforderlich. Dieses Gen ist an der Regulation der Autophagie beteiligt und spielt eine wichtige Rolle bei Entwicklung, Tumorgenese und Neurodegeneration. Schizophrenie ist mit niedrigen Beclin-1-Spiegeln im Hippocampus der Betroffenen verbunden, was zu einer verminderten Autophagie führt, die wiederum zu einem erhöhten neuronalen Zelltod führt. Es wurde festgestellt, dass Beclin-1 die Autophagie in autophagiedefekten Hefen mit einer gezielten Störung von apg6/vps30 und in menschlichen MCF7-Brustkarzinomzellen fördern kann.

BECN1 (beclin 1; Beclin-1/ATG6) is a core autophagy regulator and class III PI3K-complex component linking PI3P production to autophagosome initiation and membrane trafficking.
Biological function: Beclin-1 acts as a core subunit of class III phosphatidylinositol 3-kinase complexes that generate PI3P. Distinct BECN1-containing complexes support autophagosome initiation, autophagosome maturation and endocytic trafficking, making BECN1 central to macroautophagy and membrane-remodelling pathways.
