Das Huhn IL12A(Interleukin 12A) ELISA Kit ist ein ELISA-Kit von ELK Biotechnology zur Forschungsmessung von IL12A.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem Antikörper vorbeschichtet, der spezifisch für Hühner-IL12A ist. Standards oder Proben werden den entsprechenden Vertiefungen der Mikrotiterplatte zugegeben, dann mit einem biotin-konjugierten Antikörper, der spezifisch für Hühner-IL12A ist. Anschließend wird Avidin, konjugiert mit Meerrettichperoxidase (HRP), zu jeder Mikrotiterplattenvertiefung gegeben und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur jene Vertiefungen eine Farbänderung, die Hühner-IL12A, den biotin-konjugierten Antikörper und enzymkonjugiertes Avidin enthalten. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von Hühner-IL12A in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
