Das Human MAG (Myelin Associated Glycoprotein) Microsample ELISA Kit ist ein ELK Biotechnology ELISA Kit zur Messung von MAG in der Forschung.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte ist mit einem Human-MAG-spezifischen Antikörper vorbeschichtet. Standards oder Proben werden in die entsprechenden Vertiefungen der Mikrotiterplatte gegeben, anschließend ein biotin-konjugierter Antikörper, der spezifisch für Human-MAG ist. Danach wird Avidin, das an Meerrettichperoxidase (HRP) konjugiert ist, zu jeder Mikrotiterplattenvertiefung gegeben und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur jene Vertiefungen eine Farbveränderung, die Human-MAG, den biotin-konjugierten Antikörper und das enzymkonjugierte Avidin enthalten. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und die Farbveränderung spektralphotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von Human-MAG in den Proben wird anschließend durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
