Das Maus-IL1a(Interleukin 1 Alpha) Microsample ELISA Kit ist ein ELISA-Kit von ELK Biotechnology zur Forschungsmessung von IL1a.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem Antikörper vorbeschichtet, der spezifisch für Maus-IL1a ist. Standards oder Proben werden den entsprechenden Mikrotiterplattenvertiefungen hinzugefügt, dann mit einem biotin-konjugierten Antikörper, der spezifisch für Maus-IL1a ist. Anschließend wird Avidin, das an Meerrettichperoxidase (HRP) konjugiert ist, zu jeder Mikrotiterplattenvertiefung gegeben und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur jene Vertiefungen, die Maus-IL1a, biotin-konjugierten Antikörper und enzymkonjugiertes Avidin enthalten, eine Farbänderung. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von Maus-IL1a in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
