Das Humane IL12A (Interleukin 12A) ELISA Kit ist ein ELK Biotechnology ELISA Kit für die Forschungsmessung von IL12A.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte ist mit einem Human-IL12A-spezifischen Antikörper vorbeschichtet. Standards oder Proben werden in die entsprechenden Vertiefungen der Mikrotiterplatte gegeben, danach wird ein biotin-konjugierter Antikörper, der spezifisch für humanes IL12A ist, hinzugefügt. Als Nächstes wird Avidin, das an Meerrettichperoxidase (HRP) konjugiert ist, zu jeder Mikrotiterplattenvertiefung gegeben und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur die Vertiefungen, die humanes IL12A, biotin-konjugierten Antikörper und enzymkonjugiertes Avidin enthalten, eine Farbänderung. Die Enzym-Substrat-Reaktion wird durch Zugabe einer Schwefelsäurelösung beendet, und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration des humanen IL12A in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
