Das Human MMP10 (Matrix Metalloproteinase 10) Microsample ELISA Kit ist ein ELISA-Kit von ELK Biotechnology zur Forschungsmessung von MMP10.
Das in diesem Kit angewendete Testprinzip ist ein Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem Antikörper vorbeschichtet, der spezifisch für humanes MMP10 ist. Standards oder Proben werden zu den entsprechenden Vertiefungen der Mikrotiterplatte gegeben, dann mit einem Biotin-konjugierten Antikörper, der spezifisch für humanes MMP10 ist. Anschließend wird Avidin, das an Meerrettichperoxidase (HRP) konjugiert ist, zu jeder Mikrotiterplattenvertiefung gegeben und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur diejenigen Vertiefungen eine Farbänderung, die humanes MMP10, biotin-konjugierten Antikörper und Enzym-konjugiertes Avidin enthalten. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von humanem MMP10 in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
