Der Human MIP3b (Macrophage Inflammatory Protein 3 Beta) ELISA Kit ist ein ELK Biotechnology ELISA Kit für die Forschungsmessung von MIP3b.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem Antikörper vorbeschichtet, der spezifisch für humanes MIP3b ist. Standards oder Proben werden dann zusammen mit einem biotin-konjugierten Antikörper, der spezifisch für humanes MIP3b ist, in die entsprechenden Mikrotiterplattenvertiefungen gegeben. Anschließend wird Avidin, das an Meerrettichperoxidase (HRP) konjugiert ist, zu jeder Mikrotiterplattenvertiefung gegeben und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur diejenigen Vertiefungen eine Farbänderung, die humanes MIP3b, biotin-konjugierten Antikörper und enzym-konjugiertes Avidin enthalten. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet, und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von humanem MIP3b in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
