Das Maus-CTNNb1(Catenin Beta 1)-Mikroproben-ELISA-Kit ist ein ELK Biotechnology ELISA-Kit zur Forschungsmessung von CTNNb1.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem Antikörper vorbeschichtet, der spezifisch für Maus-CTNNb1 ist. Standards oder Proben werden zu den entsprechenden Mikrotiterplattenvertiefungen hinzugefügt, dann mit einem biotin-konjugierten Antikörper, der spezifisch für Maus-CTNNb1 ist. Als Nächstes wird Avidin, konjugiert mit Meerrettichperoxidase (HRP), zu jeder Mikrotiterplattenvertiefung hinzugefügt und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur die Vertiefungen, die Maus-CTNNb1, einen biotin-konjugierten Antikörper und enzymkonjugiertes Avidin enthalten, eine Farbänderung. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von Maus-CTNNb1 in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
