Das Human DEFb2 (Defensin Beta 2) Microsample ELISA Kit ist ein ELK Biotechnology ELISA Kit zur Forschungsmessung von DEFb2.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem Antikörper vorbeschichtet, der spezifisch für humanes DEFb2 ist. Standards oder Proben werden in die entsprechenden Vertiefungen der Mikrotiterplatte gegeben, dann mit einem Biotin-konjugierten Antikörper, der spezifisch für humanes DEFb2 ist. Als Nächstes wird Avidin, das an Meerrettichperoxidase (HRP) konjugiert ist, zu jeder Mikrotiterplattenvertiefung gegeben und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur die Vertiefungen, die humanes DEFb2, Biotin-konjugierten Antikörper und Enzym-konjugiertes Avidin enthalten, einen Farbwechsel. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und der Farbwechsel wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von humanem DEFb2 in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
