Der Human-BRCA1-(Breast Cancer Susceptibility Protein 1)-ELISA-Kit ist ein ELK-Biotechnology-ELISA-Kit zur Forschungsanalyse von BRCA1.
Das in diesem Kit angewandte Testprinzip ist ein Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem Antikörper vorbeschichtet, der spezifisch für humanes BRCA1 ist. Standards oder Proben werden zu den entsprechenden Mikrotiterplattenvertiefungen gegeben, dann mit einem biotin-konjugierten Antikörper, der spezifisch für humanes BRCA1 ist. Als Nächstes wird Avidin, konjugiert an Meerrettichperoxidase (HRP), zu jeder Mikrotiterplattenvertiefung gegeben und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur jene Vertiefungen, die humanes BRCA1, biotin-konjugierten Antikörper und enzymkonjugiertes Avidin enthalten, eine Farbänderung. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration des humanen BRCA1 in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.

BRCA1 (BRCA1 DNA repair associated; breast cancer type 1 susceptibility protein) is a nuclear genome-maintenance protein and E3 ubiquitin ligase central to DNA-damage responses, double-strand-break repair and genomic stability.
Biological function: BRCA1 forms a functional heterodimer with BARD1 that has E3 ubiquitin-protein ligase activity. The complex participates in cellular responses to DNA damage and helps coordinate DNA double-strand-break repair, homologous recombination, ubiquitin signalling and maintenance of genome integrity.
