Der ELISA-Kit für humanes MAP2 (Microtubule Associated Protein 2) ist ein ELK Biotechnology ELISA-Kit zur Forschungsmessung von MAP2.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem spezifischen Antikörper gegen humanes MAP2 vorbeschichtet. Standards oder Proben werden dann zu den entsprechenden Mikrotiterplattenvertiefungen mit einem Biotin-konjugierten Antikörper spezifisch für humanes MAP2 gegeben. Als Nächstes wird Avidin, konjugiert an Meerrettichperoxidase (HRP), zu jeder Mikrotiterplattenvertiefung gegeben und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur die Vertiefungen, die humanes MAP2, Biotin-konjugierten Antikörper und Enzym-konjugiertes Avidin enthalten, eine Farbänderung. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von humanem MAP2 in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
