Der Human-PRF1 (Perforin 1) Mikrosample ELISA Kit ist ein ELISA Kit von ELK Biotechnology für die Forschungsmessung von PRF1.
Das in diesem Kit angewandte Testprinzip ist ein Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem für humanes PRF1 spezifischen Antikörper vorbeschichtet. Standards oder Proben werden den entsprechenden Vertiefungen der Mikrotiterplatte hinzugefügt, anschließend ein biotin-konjugierter Antikörper, der spezifisch für humanes PRF1 ist. Danach wird Avidin, konjugiert an Meerrettichperoxidase (HRP), zu jeder Mikrotiterplattenvertiefung gegeben und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur die Vertiefungen, die humanes PRF1, den biotin-konjugierten Antikörper und das enzymkonjugierte Avidin enthalten, eine Farbänderung. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und die Farbänderung spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von humanem PRF1 in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
