Der Human-MYC(V-Myc Myelocytomatose-virales Onkogen-Homolog)-ELISA-Kit ist ein ELISA-Kit von ELK Biotechnology zur Forschungsmessung von MYC.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte ist mit einem Antikörper vorbeschichtet, der spezifisch für humanes MYC ist. Standards oder Proben werden in die entsprechenden Mikrotiterplattenvertiefungen gegeben, gefolgt von einem Biotin-konjugierten Antikörper, der spezifisch für humanes MYC ist. Anschließend wird Avidin, konjugiert mit Meerrettichperoxidase (HRP), zu jeder Mikrotiterplattenvertiefung gegeben und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur die Vertiefungen, die humanes MYC, Biotin-konjugierten Antikörper und Enzym-konjugiertes Avidin enthalten, eine Farbänderung. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet, und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von humanem MYC in den Proben wird dann durch Vergleich der OD-Werte der Proben mit der Standardkurve bestimmt.
