Das Human-GAD2(Glutamat-Decarboxylase 2, Säure)-ELISA-Kit ist ein ELK-Biotechnologie-ELISA-Kit zur Forschungs-Messung von GAD2.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem Antikörper vorbeschichtet, der spezifisch für menschliches GAD2 ist. Standards oder Proben werden zu den entsprechenden Mikrotiterplattenvertiefungen hinzugefügt, anschließend mit einem Biotin-konjugierten Antikörper, der spezifisch für menschliches GAD2 ist. Als Nächstes wird Avidin, konjugiert an Meerrettichperoxidase (HRP), zu jeder Mikrotiterplattenvertiefung hinzugefügt und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur jene Vertiefungen eine Farbänderung, die menschliches GAD2, Biotin-konjugierten Antikörper und Enzym-konjugiertes Avidin enthalten. Die Enzym-Substrat-Reaktion wird durch Zugabe einer Schwefelsäurelösung beendet, und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von menschlichem GAD2 in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
