Das Human-DNM2(Dynamin 2)-ELISA-Kit ist ein ELK Biotechnology ELISA-Kit zur Forschungsmessung von DNM2.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem Antikörper vorbeschichtet, der spezifisch für humanes DNM2 ist. Standards oder Proben werden den entsprechenden Mikrotiterplattenvertiefungen hinzugefügt, dann mit einem Biotin-konjugierten Antikörper, der spezifisch für humanes DNM2 ist. Als Nächstes wird Avidin, konjugiert an Meerrettichperoxidase (HRP), jeder Mikrotiterplattenvertiefung hinzugefügt und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur die Vertiefungen, die humanes DNM2, Biotin-konjugierten Antikörper und Enzym-konjugiertes Avidin enthalten, eine Farbveränderung. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und die Farbveränderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von humanem DNM2 in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
