Das Maus-AST(Aspartat-Aminotransferase)-Mikroproben-ELISA-Kit ist ein ELK Biotechnology ELISA-Kit zur Forschungs-Messung von AST.
Das in diesem Kit angewendete Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem Antikörper vorbeschichtet, der spezifisch für Maus-AST ist. Standards oder Proben werden den entsprechenden Mikrotiterplattenvertiefungen hinzugefügt, gefolgt von einem biotin-konjugierten Antikörper, der spezifisch für Maus-AST ist. Anschließend wird Avidin, konjugiert mit Meerrettichperoxidase (HRP), jeder Mikrotiterplattenvertiefung hinzugefügt und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur jene Vertiefungen, die Maus-AST, biotin-konjugierten Antikörper und enzymkonjugiertes Avidin enthalten, einen Farbwechsel. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet, und der Farbwechsel wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von Maus-AST in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
